In the past three years of my postdoc, I established a brain organoid lab from scratch. I organized documentation for the GMO S1 and S2 lab work. I optimized guided and unguided brain organoid protocols for human and non-human primate cell lines. To overcome the heterogeneity of organoids, I established mid-throughput quality control pipelines. I charactreized the transcriptional profile of generated organoids with high-throughput snRNAseq and ATACseq. I also performed functional validations via CRISPR knockout experiments in human iPSCs.

I worked with human-specific endogenous retrovirus, studying its role in the genome protection of human embryonic stem cells. I performed a wide range of RNA interference and CRISPR experiments, optimised PCR assays for challenging repetitive sequences and perfromed luciferase assays.  

Skills

Laboratory setup: S2 organoid lab infrastructure, SOPs/manuals, QC workflows, cell line acquisition and banking, GMO project documentation for regulatory approval. Organoids and cell culture: Human and primate iPSC-derived cortical and cerebellar organoids, organoid transfections, cryosectioning, wide-field/confocal microscopy, whole-organoid imaging. Gene editing: Plasmid cloning, CRISPR/Cas9 and Cas13 editing, shRNA knockdown, stem cell transfection/electroporation, AAV design and production. Molecular assays: Advanced PCR for challenging templates, nested/gradient/touchdown PCR, gene expression and protein assays, RNA-protein immunoprecipitation, luciferase assays. NGS: 10X snRNA-seq and scATAC-seq sample preparation, multiplexed snRNA-seq, bulk and circRNA-enriched library preparation. Data analysis: R, command line, ImageJ/Fiji. Teaching: Hands-on practicum training and supervision of thesis students.
